Betanodavirus infections cause significant mortality in over 30 marine fin-fish species, with clinical disease generally occurring during early fry and fingerling stages. The ensuing disease has been termed VNN (viral nervous necrosis) or VER (viral encephalopathy and retinopathy). Currently, primary virus isolation for the ACNNV routinely occurs according to OIE protocols in the SSN-1 cell line, a mixed cell culture line containing a C-type retrovirus derived from Striped Snakehead. In low titre infections virus isolation and confirmation can take over 10 days. Work completed by Iwamoto and colleague in 2000 resulted in the development of a clone of the SSN-1 which they termed E-11. Subsequent work by these researchers in 2001 confirmed that detection of Betanodavirus could be improved using a combination of cultivation in the E-11 cell line followed by a RT-PCR assay after only 48 hours incubation. These protocols were applied as a means to more rapidly isolate and confirm ACNNV infection during routine diagnostic testing. We report here a description and characterization of C-type retrovirus harbored by the E-11 cell line, comparison of the sensitivity of SSN-1 and E-11 cell line for isolation of the ACNNV, provide an improved diagnostic protocol for isolation and confirmation of ACNNV and potentially all Betanodaviruses.
