Virus; procedure to detect virus by using GLIFT Kit
Complete grinding the small piece of tissue of potato (≈2x2 cm) in plastic bag with 1 ml of extraction buffer (Use 0.5-1 more ml of extraction buffer to dilute their sticky crude extraction)
Put 3 drops of crude extraction of sample from No.1 in to well of GLIFT Kit check the reaction in 3-5 min.

Bacteria; procedure to detect Ralstonia solanacearum by using GLIFT Kit
To confirm that seed potatoes are R. solanacearum-free, an efficient detection method is necessary. Several methods can be used for plant pathogenic bacteria diagnosis. The diagnosis from infested tissue by culturing on the media requires 5-7 days. Faster diagnosis methods such as Enzyme-linked Immunosorbent Assay (ELISA) or Dot Immunobinding Assay (DIBA) can be used to identify plant pathogenic bacteria because of their accuracy, quick and large number of samples can be tested at the same time, but these methods require approximately 5-6 hours.
The ELISA test kit for R. solanacearum detection has been developed; however, this kit requires user’s skill and experience which limits the utilization in the fields. A more user friendly, the Gold labeled IgG flow test (GLIFT), kit was developed using serology technique in combination with Lateral flow test technique. It has high accuracy, precision and fast. The result could be obtained within 5-10 minutes regardless of user’s experience and skill. The GLIFT kit can be used for R. solanacearum testing in bacterial wilt-free certification program. The results obtained from the GLIFT kit were proved to be consistent with the laboratory results. Farmers can use the kit to test for seed potatoes prior to planting to reduce the disease in the field. The GLIFT kit can detect as low as 104 cfu/ml of R. solanacearum in seed potatoes sap, the violet color appears in the test line and the control line within 5 minutes. The bacteria detection

The high temperature and unsuitable fertilizer ratio are affected to impact crops. Moreover, potato virus are seed-borne that it is reduce mass production of seed potato. Then, the technique of in vitro micropropagation in potato cv. Atlantic will be done. The technique of induce microtuber in solid and liquid media should be develop. The minituber production under aeroponic system should be improves technique, lower costs and increase the farmers’ income. Aeroponics is a soilless method for producing pre basic potato seed. The method can produce higher yields, easier, rapid, flexible and more security for the phytosanitary quality.
