BASIC DISPENSING THEORY
METHODS OF MANUFACTURING TABLETS
There are 3 methods of producing tablets
Direct compression
Dry granulation
Wet granulation


DIRECT COMPRESSION: This involves mixing the drugs and all the additives and compressing them into tablets. The advantages include:
The method is very quick
Simple and
Economical.
Heat is not employed,no addition of liquids.This method is suitable for water and heat-sensitive drugs.
There is high dissolution rate

The disadvantages include:


1.It is only limited to low-dose drugs
2.Segregation is likely to occur when the active drug have a different particle size from the other ingredients.
PROPERTIES OF A GOOD DIRECT COMPRESSION EXIPIENT
1. It should have good flow properties
2. It should have good compressibility

Common direct compression exipients are: microcrystalline cellulose, spray dry lactose, hydrolysed starch, dicalciumphosphate. E.g. of drugs that can be directly compressed are: Aspirins.


DRY GRANULATION
ADVANTAGES
It is suitable for water and heat sensitive drugs
Avoids the problem of separation of exipients that have different particle sizes or densities
It ia suitable for fine powders


DISADVANTAGES
Since it involves double lubrication and compression,the tables may be hard which may lead to long disintegration time hence long/slow dissolution rate.
Some ingredients may loose their compressibility and properties when required to be compressed twice.


WET GRANULATION METHOD
This is the most widely used method
ADVANTAGES
It can be used for exipients with different particle sizes and densities that cannot be compressed by the method of direct compression.
If the drug is water sensitive,alcohol is used as granulating fluid therefore suitable for drugs that are water sensitive


DISADVANTAGES
It is time consuming and has several steps to be followed
It is not suitable for heat sensitive drugs
There may be variation in the sizes of the granules.
