Kuraklık Stresinin Kasnak Meşesi (Quercus vulcanica) [Boiss & Heldr. ex] Kotschy ve Boz Pırnal Meşesi’nde (Quercus aucheri) [Jaub. & Spach] Karşılaştırmalı Olarak İncelenmesi

Kuraklık stresi bitki büyümesini ve verimini etkileyen en yaygın abiyotik stres faktörlerinden biridir ve bitkiler yaşamları boyunca en az bir kez susuzluk dönemi ile karşılaşabilirler. Kuraklık stresinin kaçınılmaz sonuçlarından biri hücresel ve foto-oksidatif hasarı indükleyen reaktif oksijen türlerinin farklı hücresel bölümlerde (kloroplast, peroksizom ve mitokondri) üretiminin artmasıdır. Artan reaktif oksijen türlerinin çok yönlü ve işbirliği içinde bulunan antioksidan sistemler tarafından kontrol altında tutulmasıyla hücrelerarası konsantrasyonu düzenlenir. Bu nedenle yapılan bu çalışmanın amacı, Türkiye için endemik olan Quercus vulcanica ve Quercus aucheri’nin yaprak ve gövdelerinde, kuraklık stresinin fizyolojik ve antioksidan sistemler üzerindeki etkilerinin incelenmesidir. 6 aylık meşe fideleri 6 farklı deney grubuna ayrılarak 1, 2 ve 4 haftalık kuraklık stresine tabi tutuldular. 1, 2 ve 4 haftalık kuraklık uygulaması sonucunda i) taze ve kuru ağırlık, fotosentetik pigment, prolin, antosiyanin ve total çözünebilir protein içerikleri, ii) lipid peroksidasyon, indirgenmiş ve okside glutatyon, askorbik asit içerikleri ve süperoksit dismütaz, katalaz, askorbat peroksidaz, guaiakol peroksidaz ve glutatyon redüktaz gibi antioksidan enzimlerin aktiviteleri Quercus vulcanica ve Quercus aucheri’nin yaprak ve gövdelerinde tayin edildi.

1, 2 ve 4 haftalık kuraklık stresi sonucunda Quercus türlerinde taze ve kuru ağırlık içerikleri kontrol grubuna göre önemli ölçüde azalmış olup, klorofil içerikleri azalırken, karotenoit, prolin ve antosiyanin içerikleri ise artmıştır. Askorbik asit, indirgenmiş glutatyon ve total çözünebilir protein içerikleri Quercus türlerinin yapraklarında azalırken gövdelerinde artmış olup, okside glutatyon içeriği ise hem yapraklarda hem de gövdelerde artmıştır. Kuraklık stresi altındaki Quercus türlerinin yaprak ve gövdelerinde lipid peroksidasyon ve hidrojen peroksit seviyeleri artmış olup, bu da antioksidan enzim aktivitelerinde bir takım değişikliklere sebep olmuştur. Sonuç olarak, fizyolojik ve biyokimyasal bulgular Quercus vulcanica ve Quercus aucheri’nin yaprak ve gövdelerinde kuraklık stresinin lipid peroksidasyonunu indüklediği ve fizyolojik ve antioksidan sistemlerde de bir takım değişikliklere sebep olduğunu göstermiştir. Yapılan bu çalışma kuraklık stresi koşulları altında yetiştirilen ve Türkiye için endemik olan Quercus türleri üzerindeki gelecekteki çalışmalara katkıda bulunacaktır.  

 

 



A Comparative Study of Drought Stress on Kasnak Oak (Quercus vulcanica) [Boiss & Heldr. ex] Kotschy and Boz Pırnal Oak (Quercus aucheri) [Jaub. & Spach]

Drought stress is one of the most common factors of abiotic stress which affects plant growth and productivity world-wide and plants are likely to encounter periods of water shortage at least once in their lifecycle. One of the inevitable consequences of drought stress is enhanced reactive oxygen species production which induces cellular damage and photo-oxidative damage in the different cellular compartments, namely in the chloroplasts, the peroxisomes and the mitochondria. This enhanced reactive oxygen species production is however under tight control by a versatile and cooperative antioxidant system that modulates intracellular reactive oxygen species concentration. Therefore, the aim of the present study was to investigate disturbances on physiological process and antioxidative systems in the leaves and the stems of Quercus vulcanica (Kasnak Oak) and Quercus aucheri (Boz Pırnal Oak) which are endemic for Turkey under drought stress treatments. 6 months old plants were separated to 6 experimental groups and treated with drought stress for different time periods (1, 2 and 4 weeks). The aim of this study was to investigate the effects of 1, 2 and 4 weeks drought on contents of fresh and dry weight, photosynthetic pigments, proline, anthocyanin and total soluble protein, ii) on lipid peroxidation, reduced and oxidized glutathione, ascorbic acid, and antioxidant enzymes such as superoxide dismutase, catalase, ascorbate peroxidase, guaicol peroxidase, glutathione reductase in the leaves and the stems of Quercus vulcanica and Quercus aucheri.

Our results showed that after 1, 2 and 4 weeks treatments of drought significantly decreased levels of fresh and dry weight in the leaves and the stems of Quercus species when compared to control groups. Levels of chlorophyl were found to be decreased while levels of caratenoid, proline, anthocyanin were increased. Levels of ascorbic acid, reduced glutathione and total soluble protein were found to be decreased in the leaves while increased in the stems of Quercus species and oxidised glutathione increased in both leaves and stems. Levels of lipid peroxidation and hydrogen peroxide were found to be increased, therefore the activities of antioxidant enzymes had some disturbances in the leaves and the stems of Quercus species under drought stress treatments. In conclusion, physiological and biochemical investigations demonstrated that treatments of drought results in the induction lipid peroxidation and changes in physiological process and antioxidant systems in the leaves and stems of Quercus vulcanica and Quercus aucheri. The present study would contribute to future studies on Quercus species which are endemic for Turkey under drought stress conditions.
